Urszula Hibner's team discovered a novel activity of NS5A, a nonstructural HCV protein. NS5A perturbs hepatocyte epithelial polarity and cooperates with TGF-beta and with oncogenic Ras to induce aggressive liver tumours (*Akkari, *Gregoire et al (2012). Hepatitis C viral protein NS5A induces EMT and participates in oncogenic transformation of primary hepatocyte precursors. Journal of Hepatology/57/, 1021-1028). The Institute of Molecular Genetics of Montpellier (IGMM) is a multidisciplinary institute for advanced studies in biology. It has an internationally-acknowledged reputation in the fields of genetic and epigenetic regulations, RNA biology as well as cell cycle, death and metabolism. Cyclin A2 plays a key role in cell cycle regulation both in S phase and mitosis. It is essential in embryonic cells and in the hematopoietic lineage, yet dispensable in fibroblasts and other adult tissues. In a work published in January 2012 in The Journal of Cell Biology, Nikola Arsic and collaborators demonstrate that Cyclin A2-depleted cells display a cortical distribution of actin filaments and increased migration. These defects are rescued by restoration of wild-type Cyclin A2, which directly interacts with RhoA, or by a Cyclin A2 mutant unable to associate with CDK. In vitro, Cyclin A2 potentiates the exchange activity of a RhoA-specific GEF. Consistent with this, Cyclin A2 depletion enhances migration of fibroblasts and invasiveness of transformed cells via down regulation of RhoA activity. Moreover, Cyclin A2 expression is lower in metastases relatives to primary colon adenocarcinoma in matched human tumors . All together, these data show that Cyclin A2 negatively controls cell motility by promoting RhoA activation, thus demonstrating a novel Cyclin A2 function in cytoskeletal rearrangements and cells migration.
